Karako hasn't had much of her history described in the series (if at all), so most of it is speculation. She's not on Wikipedia, either, as Deadman Wonderland is a small fandom and she is a relatively new character.

What we do know, however, is that she is one of the Deadmen locked in the Deadman Wonderland prison. That prison is in fact a cover up for experiences conducted on certain people who have been touched by the nameless worm, which allows them to use their blood as a weapon in various ways. The guards themselves know nothing of this, and the head of the organization is an old, almost dead man. Things are done in secret, and as far as the inmates know (for the few who do, and the Deadmen themselves), illegally.

Karako helps Nagi, another Deadman, to form a rebellion group to free them all from DW, in order to regain their freedom. The Deadmen are used for brawls called Corpse Carnival, in which two are forced to fight, and people pay a high price to watch these matches anonymously. The loser loses a part of his body (decided by a slot machine), and the winner gets a cash prize. Sick of being used this way, and wanting to live their own lives again, multiple Deadmen join Karako and Nagi in Scar Chain, the name of their group.

Karako refuses to tell Nagi just why she helps him out, or what freedom it is she seeks. In actuality her freedom is the power to help the people she loves, pushing them to keep going forth from behind, and keeping them safe. She wants to protect Nagi and help him accomplish his goal at any cost. For the people she loves, she puts aside her own desires, as their desire become her own.

She will go to great lengths to accomplish what she wants. She may appear cold, distant and dangerous to strangers, distrusting people at first sight. But once she introduces herself or judges you can be trusted, she becomes a kind, caring person. She's loud, laughs and eats a lot, and isn't as scary as she might have appeared. Don't test her, however, as she isn't one to be messed with. When she's serious about something, she is dead serious, and can indeed become dangerous.

She likes having the people she cares about happy, and does her best to achieve that.

Karako's Branch of Sin is that she has the ability to harden her blood on any part of her body at will and almost instantaneously. She can harden it in layers, creating something resembling a rock around her fist, giving her punches and hits a lot more power, and making them that much more dangerous. Her power remains generally defensive, but it suits her, as someone that seeks to protect others. This doesn't stop her from getting into fights, however – if provoked, she will not simply defend herself and run. Quite the opposite, she will be glad to indulge herself in a beating.
